You wouldn't be able to serve Ayr directly without a reversal, however you would be able to serve Newton-on-Ayr, one stop north.

Using the West and North London lines, several routes Exeter-Yeovil-London (either route)-WLL-NLL-then either of WCML/MML/ECML to Newcastle & Edinburgh, then Carlisle-Crewe-Bristol (multiple routes Reading/Gloucester/Severn tunnel) - Exeter
