It's a good idea - but you run into one major problem - the route from the end of the current no. 25 Reading - Cav - Son Com - Peppard Com route ends at the Unicorn in Peppard Common.It is a bit of a shame that Reading - Sonning Common - Henley has never been seen as a viable option instead of competition along the direct route. The journey time wouldn't be that uncompetitive relative to the Twyford / Wargrave route.
This means you either have to 1.) divert down Colliers Lane (a problem since it is a tiny with a large vehicles restriction) to reach the B481 and the route onwards to Greys and Henley, or 2.) or turn right onto Gallowstree Rd to get to the B481 much further south in Peppard - missing out a chunk of the northern section of the route.
Maybe Reading Buses will consider having a chat with the National Trust and extending the new ruby 28 from Henley up to Greys Court every hour.
The NT give a discount on entry for public transport use and there's nothing but a very infrequent village shopper bus up that way.
