Direct trains are always permitted.
The shortest route (+ up to 3 miles longer) is always permitted.

For mapped routes we need to check the Routeing Guide

The Routeing Guide is here:
http://www.atoc.org/about-atoc/rail-settlement-plan/routeing-guide

First we need to identify appropriate Routeing Points
Lancaster is a member of Lancaster Group
London Terminals is a member of London Group

Now we need to identify which maps we can use
BS+CS
BS+GC
CS+LM
CS+NC
LM
LM+EH+NC
LM+MH+NC
MM
MM+PS+NC
MM+PS+NW

Now you can look at those map combinations and work out which routes are permitted. The maps can be found here:-
http://www.atoc.org/clientfiles/File/maps.pdf
