As I understand it, the line from Sheffield to Stockbridge will be reopened. The line from Huddersfield to Sheffield via Barnsley is slow, bit quite well used given the hourly frequency. How difficult would it be to reopen the bit from penistone to Stockbridge?

The A616 Stocksbridge* bypass severs the route, so that would be an expensive problem to solve. I'm not aware of any other major breaches.
Much of the traffic on the existing route is for Meadowhall, so the twenty-minute time saving by avoiding the Barnsley dogleg would only be useful for those heading to central(-ish) Sheffield.

*- it's Stocksbridge. Stockbridge is on Merseyside.

Is Stocksbridge traffic more for Sheffield or Meadowhall?

At present it'll all be on bus or bus+tram, so I'm not sure. My previous point was based on what happens with the Huddersfield trains, which empty out significantly at Meadowhall Interchange.
