My iPhone Network Rail app gives me journeys from Southampton to Waterloo via Reading and Paddington. Is this a valid routing for a London Terminals to Southampton off peak day return?

Southampton to Paddington via Reading is a Permitted Route for the return leg of a London Terminals - Southampton Central Off-Peak Day Return, but the journey has been completed on arrival at Paddington, and if you want to continue by tube to Waterloo you then need another ticket or to use Oyster.

Right. If you want to get to Waterloo I think you would need to get on an SWT service at Reading to Waterloo via Ascot.
