Your mention of a "Manchester X43" service makes me wonder if you are referring to the Transdev Burnley and Pendle "The Witch Way" X43 service that runs Nelson/Burnley to Manchester.
Stagecoach Manchester run a 43 service from Manchester to Manchester Airport that seems to run most hours.

This service must have been going for years. As a kid in the 60s I can remember Ribble double-deckers operating it.

I wouldn't expect it to be overly busy on a Saturday morning out and not full on a Sunday afternoon in, as they aren't really the times of most demand for the service. Mornings into Manchester and evenings out definitely do experience high demand.

Not on the X43, no. For many years it went via Bury New Road to Whitefield, Bury and up to Rossendale. Once the M66 opened the route split into two variants, one via the old route and one going straight up the M66. The modern X43 only has the M66 route.
Do you have google maps on your phone? I find that useful for working out when I'm nearest where I'm going when there's no other information. At worst I usually end up out by one stop,
You could also try asking the driver if he could give you a shout when you get there if there's something obvious near the stop you're heading for. Burnley and Pendle's drivers are mostly friendly, but there is a risk they might forget.

According to the timetable the X43 calls at Prestwich (Premier Inn) and then runs via the motorway to Bank Street, so it's the first stop after a non-stop run of 25 minutes on the M66 motorway.

It may be a bit late now, but I would recommend the Bus Scout app for Android as you can track any bus route following each bus stop.
Unfortunately, if you've been reading recent bus threads, many companies, even supposedly 'quality' services like the X43, don't seem to think having information on the next stop is important.
