No difference whatsoever. 66/3 was the fastline fleet and 66/4 are the original DRS fleet.
The 66/4s are/were a mixture to, as are the 66/5s.Those are, I believe, the low-emissions variant (as opposed t the 66/0s which aren't).
It seems to have been further complicated with GBRF as those are all numbered 667xx but appear to be a mixture of the two types.

The first five Freightliner 66s (501-505) were nothing to do with Heavy Haul: they were ordered for the Intermodal business. The first Heavy Haul business locos were 506-520, ordered to support a contract to provide haulage for Railtrack engineering services.Freightliner 95 Ltd created a Freightliner Heavy Haul sister company and ordered five 66/5 from EMD. I believe these five were taken from the middle of the EWS build at the London Ontario factory, and I think they were from the series around EWS 66165 ish, obviously with the balance supplied "back to" EWS. I guess the builder numbers would confirm this from the EMD JT42CWR classifications.
