Title says it all for this one.

I'll start off with Belper and Duffield having no regular service to Chesterfield. It is necessary to double back at Derby for most of the day.

Also, Leyland and WBQ is a strange one because I actually knew 3 people who lived in and around Leyland when I worked in Warrington, whose commute could have been much easier.

Croy to Camelon (and vice-versa). These two stations used to have a direct service several years ago (they are on the line between Glasgow Queen Street and Falkirk Grahamston) but the Scotrail service is now routed via Cumbernauld and not via Croy.

Another one in Scotland. Linlithgow to Dalmeny (via Winchburgh Junction). There is a midweek evening Scotrail working from Glasgow Queen Street to/from Dundee routed via Linlithgow and then over the Forth Rail Bridge, but the next stop after Linlithgow is at Inverkeithing, not Dalmeny. Also (in reverse) on its way back.

Camden Road - South Hampstead , Hasn't had a regular service since the services from Broad Street via Primrose Hill ceased , however sporadically gets regular service during engineering works when the line via Kentish Town West is closed.

Upper Holloway - West Hampstead Thameslink via Carlton road junction , Don't think this has ever had a regular passenger service using it.

The obvious one - there is one train from Newark to Retford each day, at 0710 in the morning.

There are four from Retford to Newark, but none between 0811 and 2210.

Passengers must double back from Doncaster, or go via Lincoln.

Very pedantically there have been times more recently when there have been Gospel Oak to Stratford services as well as the Primrose Hill services during engineering work…
And also very pedantically (but less so), the services went from Liverpool Street, not Broad Street, at the end.

I’d also nominate Camden Road and Finsbury Park, as well as Crouch Hill and Hornsey (I think it is).

Melton Mowbray to Syston has one train in one direction only that calls only after initially passing through the station non stop on it's way to Leicester - I think that is nice and unusual

As a National Rail service would never be as frequent as the Metrolink between Deansgate and Victoria (up to 20 trams per hour) I really do not see the need for this.
Any longer distance connections from the south wanting to use trains only can currently change at Manchester Piccadilly, those from Warrington Central at Manchester Oxford Road and most stations from the west/north at Salford Crescent or Oxford Road.
I would like to see a second TransPennine service linking Piccadilly to Victoria though and would consider this far more useful than something like the Southport on the Castlefield corridor as it would significantly improve connections from south of Manchester to the Victoria served routes.

Sorry, re reading your post I misinterpreted it as you asking if a valid argument could be made for introducing a direct service between the stations. I now realise you meant could a valid argument be made for their inclusion as not currently having direct services.

You are right: the 16.31 Barnstaple to Axminster misses St James’ Park. But that does mean no trains at all between St James’ Park and Pinhoe.
