Does anyone know if it is possible to purchase tickets for this service in advance? If so, from where?

I have tried the sites for Tajik and Uzbek Railways without success.

IF noone appears here with recent details, then I suggest you use the excellent-for these areas of this world-forum on caravanistan.com. It also has info on travelling by train there and even lists a few resellers able to handle requests from far abroad for such demands.
Probably needless to say it may also have plenty of info on other ´stan´ countries around and even some adventurous travel-blogs.

I would highly doubt you can purchase such tickets online. Just turn up at the station and buy it. Last time i travelled there i bought my tickets at the station on the day with no issues.
